Hi All,

I have a pure pitch of lacto, and was hoping to propagate and freeze a few vials. Has anybody done this successfully? Should I do an anaerobic starter? How about crashing lacto out of suspension? Freeze with glycerol?

Thanks
James

Hi All,

I have a pure pitch of lacto, and was hoping to propagate and freeze a few vials. Has anybody done this successfully?

Yes, but not for a long time. I am currently playing with techniques to make this easier in small scale. Propagation is best done in Mann Rogosa Sharp or tomato juice broth but I am playing with grape juice as a substitute.

Should I do an anaerobic starter?

Yes, surprisingly easy these days using oxygen absorbers. Best source is paranoid Septics.

Hi All,How about crashing lacto out of suspension?

Very hard to get them out of suspension: sterile filtration is the best option but hard to get right for amateurs. Fortunately this step is probably unnecessary.
